Clascoterone is an androgen receptor inhibitor developed as a cream and solution for the treatment of androgen-dependent skin disorders, including androgenetic alopecia and acne vulgaris. Clascoterone powder is a chemical substance with the chemical name 17- (1-oxopropyl) -17-hydroxyandrost-4-, 16-en-3-one. White or off white crystalline powder, molecular formula C24H34O5, CAS 19608-29-8. Very soluble in dichloromethane, freely soluble in methanol, practically insoluble in water.

Clascoterone is an antiandrogen medication which is used topically in the treatment of acne. It is also under development in a higher concentration for the treatment of androgen-dependent scalp hair loss, clascoterone exerts antiandrogenic effects by working as an antagonist at androgen receptors (ARs) expressed throughout the skin, including sebaceous glands, sebocytes, and dermal papilla cells. Clascoterone blocks the effects of testosterone and dihydrotestosterone (DHT), which are androgens that bind to the ARs and contribute to the development of androgen-dependent conditions such as acne and alopecia.

Clascoterone (cortexolone 17α-propionate, CB-03-01) is a novel antagonist of androgen receptors. It binds to androgen receptors with high affinity. By competing with androgens for binding to androgen receptors, clascoterone works by blocking the androgen receptor signalling cascades that promote acne pathogenesis, such as sebaceous gland proliferation, excess sebum production, and inflammatory pathways.

Cortexolone 17α-propionate (developmental code name CB-03-01), or 11-deoxycortisol 17α-propionate, is a synthetic steroidal antiandrogen–specifically, an androgen receptor antagonist – which is used as a topical medication in the treatment of androgen-dependent conditions including acne vulgaris and androgenic alopecia (male-pattern hair loss). It is the C17α propionate ester of 11-deoxycortisol (cortexolone); C17α esters of 11-deoxycortisol were unexpectedly found to possess antiandrogen activity, and cortexolone 17α-propionate was selected for development based on its optimum drug profile.
