Paclitaxel is a natural product with antitumor activity. TAXOL (paclitaxel) is obtained via a semi-synthetic process from Taxus baccata. The chemical name for paclitaxel is 5β,20-Epoxy-l,2α,4,7β,10β,13α-hexahydroxytax-l l-en-9-one 4,10-diacetate 2-benzoate 13-ester with (2R,3S)-N-benzoyl-3-phenylisoserine.
Paclitaxel is a white to off-white crystalline powder with the empirical formula C47H51NO14 and a molecular weight of 853.9. It is highly lipophilic, insoluble in water, and melts at around 216-217° C.
Paclitaxel is a hydrophobic mitotic inhibitor with a powerful anti-cancer effect. It belongs to the group of taxanes (together with docetaxel), which are potent cytotoxic diterpenes derived from yew trees. Paclitaxel is active against a spectrum of malignancies that are generally considered to be refractory to conventional chemotherapy.
In recent years its activity was studied extensively by a vast number of clinical trials and the drug has become an irreplaceable part of various chemotherapeutic regimens. Paclitaxel inhibits cell division and chromosome segregation by targeting tubulin and stabilizing microtubules.
Paclitaxel is a monomer diterpene extracted from the bark of taxus, a natural plant. It is a complex secondary metabolite and the only known drug that can promote and stabilize the polymerized microtubules.Isotopic tracers indicate that paclitaxel binds only to the polymerized tubules and does not react with the unpolymerized tubuloprotein dimer. Cells exposed to paclitaxel accumulate a large number of microtubules, which interfere with the cell's various functions, especially by stopping cell division at the mitotic stage and preventing normal cell division.Through the II-III clinical research, paclitaxel is mainly suitable for ovarian cancer and breast cancer, to lung cancer, colorectal cancer, melanoma, head and neck cancer, lymphoma, brain tumors also has certain curative effect.
It has a good effect on refractory ovarian cancer and breast cancer with drug resistance such as ovarian cancer and platinum, and a good prospect for the treatment of prostate cancer, head and neck cancer, esophageal cancer, germ cell tumor, endometrial cancer, lymphoma, bladder cancer, upper digestive tract cancer, small cell and non-small cell lung cancer.
